L-Tryptophan
The trials are small and short. A 2022 meta-analysis that could pool only 4 trials (Sutanto et al.) found shorter time awake after falling asleep, larger at 1g/day or more, and no effect on other sleep measures. In a 3-week trial in 57 adults with chronic insomnia, tryptophan taken with carbohydrate improved sleep while carbohydrate alone did not (Hudson et al., 2005; the dose is not in the abstract). For mood, a 2021 review found differences from placebo in 4 of 11 RCTs in healthy adults (Kikuchi et al.). No trial found a cognitive benefit in healthy adults, and a 2010 review found that tryptophan loading slows reaction time. Funding could not be checked for most trials. The 1989 eosinophilia-myalgia syndrome (EMS) epidemic was traced to tryptophan from a single manufacturer.
Evidence reviewed:
Mechanism of action
L-tryptophan is an essential amino acid. The body uses it to make serotonin, and from serotonin melatonin; it also feeds the kynurenine pathway. It is not the same as 5-HTP (5-hydroxytryptophan), tryptophan-rich foods, protein hydrolysates or alpha-lactalbumin, whose trials are not evidence for L-tryptophan supplements. In trials, effects often depended on genotype or baseline state, and a review found that tryptophan loading consistently slowed reaction time, which it attributes to mild sedation.

Human effect matrix
Based on human clinical trials only. Animal and in-vitro data excluded.
| Effect | Evidence | Magnitude | Studies |
|---|---|---|---|
| Sleep Quality | preliminary | Small | 2 |
| Mood (Healthy Adults) | mixed | Small | 1 |
| Cognition & Reaction Time | preliminary | Negligible | 1 |
Evidence key: Strong = multiple consistent RCTs · Moderate = smaller/fewer RCTs · Preliminary = early trials or small n · Mixed = conflicting results
Documented benefits
- Shorter time awake after falling asleep at 1g/day or more (a meta-analysis of 4 trials)
- Better insomnia measures in one 3-week trial in adults with chronic insomnia
- Mood differences from placebo in 4 of 11 RCTs in healthy adults (a 2021 review)
Side effects & cautions
- Eosinophilia-myalgia syndrome (EMS): the 1989 epidemic was traced to tryptophan from a single manufacturer (98% of Oregon cases took it, versus 30–48% of controls), pointing to a contaminant or production change rather than tryptophan itself. In one exposed cohort, 19% of people with definite EMS died during follow-up
- Serotonin syndrome: a case report describes it after tryptophan was combined with an MAOI and clomipramine. Starting two or more serotonergic drugs is the usual trigger, and MAOIs combined with serotonergic agents can be fatal; ask a doctor before combining tryptophan with SSRIs, SNRIs, MAOIs, linezolid, triptans or tramadol
- Slower reaction times after tryptophan loading, attributed to mild sedation
- No safety data for pregnancy, breastfeeding, liver or kidney disease were reviewed

Stacking recommendations
Ingredients that pair well with L-Tryptophan and why.
Both were in a 3-day blend trial of 16 healthy adults (with glycine, tart cherry powder and L-theanine) that shortened sleep onset (Langan-Evans 2023); the abstract gives no doses, so what tryptophan adds is unknown.
Both were in the same 3-day, 16-person blend trial (Langan-Evans 2023); we cite no trial of the pair alone.
Frequently asked questions
Is L-tryptophan the same as 5-HTP?
No. 5-HTP (5-hydroxytryptophan) is a different compound, and its trials are not evidence for L-tryptophan. Tryptophan-rich foods, alpha-lactalbumin and protein hydrolysates are different again.
Do doses under 1g a day help sleep?
That has not been shown. No placebo-controlled trial of tryptophan alone below 1g/day was found, and the sleep meta-analysis found its larger effect at 1g/day or more. The one blend trial that included tryptophan (16 people, 3 days, with glycine, magnesium, tart cherry and L-theanine) gave no ingredient doses and cannot isolate tryptophan.
What is eosinophilia-myalgia syndrome?
EMS is a serious illness behind a 1989 epidemic that was traced to tryptophan from a single manufacturer: 98% of Oregon cases had taken that product, versus 30–48% of controls, which points to a contaminant or production change rather than tryptophan itself. In one exposed cohort, 19% of people with definite EMS died during follow-up.
Can I take tryptophan with antidepressants?
Ask a doctor first. Serotonin syndrome has been reported when tryptophan was combined with an MAOI and clomipramine. Starting two or more serotonergic drugs is the usual trigger, and MAOIs combined with serotonergic agents can be fatal. We found no trial that measured the risk at low tryptophan doses.
Will tryptophan make me sharper?
No trial found a cognitive benefit in healthy adults. A 2010 review found that tryptophan loading consistently slowed reaction time, which it attributes to mild sedation.
